Research on Rapid Detection Method of Probiotic Activity Based on Respiratory Metabolic Characteristics

Abstract: Probiotic activityisa keyindicator forevaluating its eficacy.Traditional plate counting methods onlyreflect the number of viable bacteria and cannot reflect the metabolic activityof the bacteria. Microorganisms produce CO 2 during metabolism,and the amount produced is directlyrelated to bacterial activity. Gas chromatography was used to detect the CO2 concentration in the probiotic metabolism, and a standard curve was constructed to achieve quantitative analysis of activity,with μg⋅CO2⋅g-1⋅d-1 as the unit of activity evaluation. The method was validated on Lactobacillus acidophilus LA85 powderand Pediococcus acidilactici PA53 granules.Theresults show thatthis method can effectively distinguish the activity differencesbetween diferent batches of products,withadetection limit of O.O05 ppm((V/V) CO2 .Compared with traditional methods,this method has theadvantages of simple operation,low cost,and accurate results, providing a new technical means for probiotic quality control.
