Improvement of UPLC-MS/MS method for determining mphenicol Residues in aquaculture water

Abstract:A detection method for amphenicol antibiotic residues,chloramphenicol,methicilin,and florfenicol residues in aquaculture water based on rough active sampler-solid phase extraction- UPLC-MS/MS was improved. After filtration,the aquaculture water sample was filtered and cleaned up by Phenomenex SPE,eluted by 5mL methanol at a rate of 1.0mL/min ,dried by nitrogen gas and redissolved. The separation was performed using Waters ACQUTY UPLC HSS C18 ( 100mm×2.1 mm i. d., 1.8μm ) in gradient elution with methanol-water as the mobile phase. The samples were detected by multiple reaction monitoring (MRM) ESI- mode and quantified by internal standard method. Chloramphenicol and methicillin achieved a good linearity in the range of 0.2~10ng/mL the linearity of florfenicol was in range of 0.1~5.0ng/mL ,with a correlation coefficient r2>0. 99 : The detection limit of chloramphenicol and methicillin in actual spiked samples was 0.2ng/L ,the quantification limit was 0.5ng/L ,and the average recoveries range was between 81.2% and 94.7% : The detection limit of florfenicol in actual spiked samples was 0.1ng/L and a quantification limit of 0.25ng/L , with average recoveries range between 84.5% and 95.7% . This method has high accuracy and short detection time,and is suitable for simultaneous determination of chloramphenicol,methicillin,and florfenicol in aquaculture water.
