Effects of Different Hormone Ratios on In Vitro Growth and Propagation Efficiency of Malus prunifolia Plantlets

Abstract:【Objective】To optimize the invitrorapid propagation system for Malus prunifolia fromWuqi,this study sys tematically investigated the regulatory effects of 6- Benzylaminopurine (6-BA) and auxin(IBA) concentration ratios in MS medium on the growth characteristics,propagationeficiency,and physiological indicators of tube plantlets.【Methods】A two-factorcompletelyrandomized design was employed,withfiveconcentrationsof6—BA(O.4,0.5,0.6,0.7,0.8mg/L) and five concentrations of IBA(0.5,0. 6,0.7,0.8, 0.9mg/L) ,forming lO treatment combinations ( T1-1 toT ). A hormone-free MS medium served as the control (CK).After 30 days of culture,parameters such as propagation coeficient, plant height,stem diameter,internode length,and chlorophyllcontent were measured. 【Results】The ratio of 6- BA to IBA significantly influenced the growth of tube plantlets. The combination of 0.7mg/L 6-BAand 0.6mg/L IBA (T5-2) )yielded the highest propagation coefficient (14.125),while 0.5mg/L 6-BA and 0.9mg/L IBA (T5-1) resulted in the best performance in stem diameter and chlorophyllcontent.【Conclusion】Principal component analysis indicated that a 6- BA/IBA ratiobetween1.Oand1.2(e.g.,.17)synergisticallyenhanedproiferationeficiencyandphysilogicalindicators,fectively addressing the issue of excessive elongation (internode length
