Development validation of quantitativemethod for ergosterol inAcremoniumterricolaculture

AbstractTo establish anaccurate reliable method for the quantitative analysis of ergosterol in Acremonium tericolaculture,thisstudyoptimizedthedetection wavelength,extractiontime(15,25,35,45,55min),extraction solvent (methanol,absolute ethanol, 95% ethanol),mobile phase ratio (methanol:water,100:0,98:2,95:5,90:10), flow rate (0.6,0.8,1.0,1.2,1.4mL/min)basedon the conventional high-performance liquid chromatography (HPLC), subsequentlyvalidatedtheoptimizedmethodintermsofspecificity,linearityprecision,accuracylimitofdetection (LOD) limitof quantification (LOQ),verifiedthe aplicabilityof the method using 10 batches of Acremonium tericola culture samples;theresultsshowed that the optimal conditions of the established HPLC method were as follows: detection wavelength at 281 nm, extraction solvent of 95% ethanol, extraction time of 45 min, mobile phase ratio of methanol to water at 100÷0 , flow rate of 1.O mL/min,this method could effectively separate ergosterol from other components inAcremonium tericola culturewithout interfering with thedeterminationresultsof ergosterol,ergosterol exhibited a good linear relationship in the concentration range of 0.5-40.0μg/mL ( R2=0.999 2 ), the relative stard deviation (RSD) of precision was less than 3% ,the recovery rate ranged from 90.45% to 99.41% , the LOQ was

0.50μg/ml , the LOD was 0.15μg/mL ,thedetection results of Acremonium terricola culture samples from the same batch showed goodrepeatability,indicating thatthemethod had high stabilityoperability; inconclusion,the HPLC method established in this studyhas the advantages of wide determination range,good linearity high accuracy, which can be used for the accurate quantification of ergosterol in Acremonium terricola culture.
