Screeningand functional validation of endogenous promoters from Bacillus amyloliquefaciens forfengycin synthesis

Abstract:UsingBacilusamloliquefaciens28astehost,ndbyintegratinggenomicandtranscriptoicdata,2upstrado enouspromotersfromhighlyexpressedgeneswerescreeed.ByconstructingaGFPreportersystemandevaluating theactivityofeach promoterinBacillus,itwasfoundthathepromoterstrengthsofPspoVG,PqoxC,PqoxB,andPgpdwerehigherthanthatoftecommonlyusedpromoterP43.WhendrivenbythePspoVGpromoter,thetranscriptionallevelofthefenCgenewas4.62timesthaofthe P43 promoter,and the fengycin yield increased from 84.3mg/L to 101.2mg/L .Regarding promoter structure,the sequences of the -35 and -10 regions ofPspoVG were highlyconsistent with theconsensussequences oftypicalstrong promoters,which mightbethestructural basis for its high-eficiency transcriptional drive.
