Evaluation of salt tolerance and physiological response mechanisms in Paeonia lactiflora

Abstract:Takingtwoherbaceouspeony(PaeonialactifloraPall)cultivars,LanFushi(L)andDaFugui(D),astestmaterials,a potexperimentwasconductedtoinvestigatethegrowthandphysiologicalchangesofherbaceouspeonyunderfiveNaClconcentration treatments(O,0,20,30,40mmol/L).TheresultsshowedthatthesalttoleranceofLanFushi(L)wastrongerthanatofDa Fugui(D).WiththeincreaseofNaClconcentration,thephenotypicsore,eafrelativeelectricalconductivity,solubleproteincontent,soluble sugarcontent,malondialdehyde(MDA)contentandperoxidase(POD)activityofbothcultivarsincreasedsignificantly, while the leaf thickness,net photosynthetic rate ( Pn )and maximum photochemical quantum yield ( Fv/Fm )decreased significantly. Correlationanalysisindicatedthatthesalt-tolerancephenotypicscorewassignificantlynegativelycorrelatedwithplantheight( P< (204号 0.05),and extremely significantly negatively correlated with leaf thickness,SPAD value, Pn , Fv/Fm and leaf relative water content ( P <0.01).Meanhile,twasextremelysignificantlypositivelyorelatedwithleafrelativeelectricalconductivity,solubleproteicon tent,soluble sugar content,catalase (CAT) activity,POD activity and MDA content( P<0.01 ),but had no significant correlation with superoxide dismutase(SOD) activity ( P>0.05 ).Furthermore,stepwise regression analysis was used to establish the optimal linearregressionequation:Salt-toleranephenotypicsore=4.198-3.747×F/Fm,andF/Fmasidentifidasanevaluationiexfor salt tolerance of herbaceous peony.
