Analysis of the Causal Relationship Between Blood Metabolites and Cerebral Hemorrhage
Based on Mendelian Randomization

Abstract: Objective To explore the causal relationship between serum metabolites and intracerebral hemorrhage (ICH) by two-sample bidirectional Mendelian randomization (MR) analysis. Methods Genome-wide association study (GWAS) data of cerebral hemorrhage were obtained from public databases. A published GWAS containing 1400 metabolites was used as the exposure factor, and SNPs significantly related to the exposure factor were selected as the instrumental variable (IV). R software was used to analyze the causal relationship between serum metabolites and cerebral hemorrhage. The methods used include inverse variance weighting (IVW), MR–Eggeer regression and simple model method. The data results were analyzed such as heterogeneity, pleiotropy, sensitivity, etc. to verify the reliability of the results. The results were verified by reverse MR analysis. Results Metabolite "1-stearoyl-2-arachidonic acid-gpc (18:0/20:4)" (IVW: OR=0.888, 95% CI: 0.830–0.950, P<0.001) was negatively correlated with cerebral hemorrhage. Conclusion 1-Stearoyl-2-arachidonic acid-gpc (18:0/20:4) is a protective factor for cerebral hemorrhage. With the increase of this metabolite, the risk of cerebral hemorrhage decreases. This may provide a new reference for the pathogenesis, early prevention, clinical treatment and scientific research of cerebral hemorrhage.
