Exploring the Mechanism of Poria Cocos in the Treatment of Nonalcoholic Fatty Liver Disease Based on Network Pharmacology

Abstract: Objective To explore the potential mechanism of Poria cocos in the treatment of non-alcoholic fatty liver disease (NAFLD) based on network pharmacology. Methods The effective components of Poria cocos were screened by TCMSP, TCMID database, and their targets were predicted by SwissTargetPrediction database. Human NAFLD-related genes were obtained from the GeneCards database and converted into targets through the Uniprot database. Venny2.1.0 was used to capture the intersection of drug and disease targets and to plot Venn maps. The PPI networks were constructed by the STRING database, and Cytoscape 3.7.1 was employed for mapping and core target screening. The DAVID database was used for GO functional enrichment analysis and the KEGG enrichment analysis. The drug-disease-pathway interaction network were constructed by the Cytoscape3.7.1 software. Results A total of 12 active components of Poria cocos were identified, primarily including eburicoic acid, pinicolic acid, and pachymic acid. And totally 1191 NAFLD targets and 111 common targets of Poria cocos were obtained. PPI results showed that IL-6, TNF were the core targets with high values. GO enrichment analysis involved biostimulation of reactive poisons, membrane raft. The results of KEGG enrichment were related to insulin resistance, alcoholic liver disease. Among the active components of Poria cocos, eburicoic acid ranked first in terms of degree, betweenness centrality, and closeness centrality. Conclusion Poria cocos may exert therapeutic effects against NAFLD by modulating pathways related to insulin resistance, alcoholic liver disease, and others. Eburicoic acid may play a significant role in these effects.
