Analysis of the gut bacterial diversity of bedbugs by high-throughput sequencing

Abstract To investigate the composition and diversity of gut bacteria in bedbugs, high-throughput sequencing of the V3–V4 hypervariable region of the 16S rRNA gene of gut bacteria from the Cimex hemipterus and Cimex lectularius species was performed using the Illumina NovaSeq platform. The number of amplicon sequence variants (ASVs) of gut bacteria was counted, and their species composition, alpha diversity, and beta diversity were analyzed. The results showed that 1 372–1 732 and 481–797 ASVs were obtained from C. hemipterus and C. lectularius, respectively; among them, 3 681 ASVs were unique to C. hemipterus and 1 599 ASVs were unique to C. lectularius. At the phylum level, the gut bacteria of both bedbug species were mainly composed of Proteobacteria, Actinobacteriota, and Firmicutes. At the class level, the dominant flora in the gut of C. hemipterus included Alphaproteobacteria (27.44%, 62.07%, 20.39%), while that in C. lectularius was dominated by Actinomycetes (52.90%, 82.17%, 56.79%). At the order level, Rickettsiales was the shared dominant flora between the two species. At the family level, the dominant gut flora of C. hemipterus included Rickettsiaceae (7.26%, 51.70%, 3.61%), whereas C. lectularius was dominated by Geodermatophilaceae (50.82%, 52.70%, 80.18%). At the genus level, Wolbachia was the shared dominant genus in the gut of both bedbug species, and Rhodococcus was unique to the gut bacteria of C. lectularius. Alpha diversity analysis indicated that the observed species number, Chao1 index, Shannon index, Simpson diversity index, and Pielou evenness index of C. hemipterus were all higher than those of C. lectularius. Beta diversity analysis showed that principal coordinate analysis (PCoA) explained 90.4% of the variation among samples, indicating clear separation in community composition between the two species. This study reveals divergent diversity in gut bacterial communities between C. hemipterus and C. lectularius, which provides a scientific basis for bed bug management via microbial control strategies.
