Abstract:To explore the functionof the S-adenosylmethionine synthetase (SAMS)gene,this studyamplified theopen eading frame(ORF)of S-adenosylmethioninesynthetase genefromAnoectochilus formosanus (AfSAMS)byPCR based ontranscriptome sequencing.Furthermore,bioinformaticstoolswereusedtoanalyzethephysicochemicalcharacteristicsoftheamplifedfragmet.The results showed that the ORF of AfSAMS was 1 191 bp in length, encoding a peptide composed of 396 residues. The putative protein had a molecular formula of C1898H2968N508O601S16, a molecular weight of 43 032.45 Da, a theoretical isoelectric point of 4.97,and an instabilityindex of25.39whichindicatedastableprotein.Theproteinhadanaliphaticaminoacidindex of80.96,conforming tothe structural characteristics of intracellular stable enzymes.The grand average of hydropathicity (GRAVY) was -0.246 ,which indicated a hydrophilic protein. The a -helixes, β -sheets, β -turns,and random coils in the secondary structure of theprotein accounted for 35.61% 15.15% 6.82% ,and 42.42% ,respectively. The predicted tertiary structure of the protein was highly similar to those of Dendrobium catenatum (XP_020681927.1) and Canna indica (WOK96063.1).
