Abstract:ObjectiveToinvestigatetheinvitroantioxidantand hypoglycemicactivitiesof AraliachinensisL.rotand itsprescription,providingascientificbasis fortheirpotentialapplicationinfunctional foodsordrug development.Methods The 75% ethanolextractsofraliachinensisLrotanditsprescriptiowereobainedusingulrasound-asistedextractionTetotalsapoincontentwasdeterminedbythevanilin-perchloricacidmethod.AntioxidantactivitywasevaluatedusingDPPHandABTSradicalcavenging assays,while hypoglycemic activity was assessed via α - glucosidase inhibition experiments. Results The total saponin content of Aralia chinensis L. root reached 25.64mg/g .The root extract exhibited significant antioxidant capacity,with enhanced activity observed in prescription ① (Aralia chinensis L. + Senna tora(L.)Roxb.)and prescription ② (Aralia chinensis L. + Euphorbia humifusa W.). Prescription ② demonstrated strong antioxidant effects ( 1C50 values for DPPH and ABTS radical scavenging were 29.46 and 1.54mg/mL ,respectively). In hypoglycemic tests,prescription ② showed synergistic effects,with α - glucosidase inhibitory activity ( lC50=0.38mg/mL )surpassing that of acarbose( 1C50=0.42mg/mL ).Enzyme kinetic analysis indicated that prescription ② irreversiblyinhibited α - glucosidase. Conclusion Prescription ② exhibits significant in vitro antioxidant and hypoglycemic effects, makingitapromisingandidateforfurtherdevelopmentasanaturalantioxidantorhypoglycemicfunctionalingredient.Thesefindings providetheoreticalsupportforotiizing rditioalChinesemedicineompoundfoulatiosndteirotentalaplicatinindbetes adjuvant therapy.
