Abstract:TakingtreshalltissueofAmooallusoncK.Kochsaterals,teifuencsofsoationrs centnucleiccidstains,fleringtimes,ndcentrifugationprotocolsonplidydetectionwerecompared.Theresultsshowedat,MGb lysisbuffer yielded significantly higher counts of A. konjac nucleus compared to MgSO 4 andTri s-MgCl2 buffers,witha prominent peak and minimal fragment peaksobserved.The coefficient of variation( C V)was calculated at 4.47% .Fluorescent staining with DAPI demonstratedcomparableeficacytoP,withtheaddedadvantageofreducingbackgroundfluorescencenoisewithoutRNAasetreatment, therebyconservinganalysistime.Boththeprimaryfiltrationandthesecondaryfiltrationmethodscouldefectivelyemoveadhered cellsandcelldebris.Thesecondaryfiltrationmethodcollectedmorecellnucleiandhadabettefect.Optimaluclearsuspesion preparationinvolveddirectligtavodaneandinsitusaningpost-fltratio.TheproposedflowcyometricprotocolforA.ojacploi dyanalysis involved:homogenizing 0.25g offreshcallus tissue in1mLMGblysisbuffer,filtering,stainingwith 50μL DAPI in darkness for 3O minutes,followed by immediate analysis.
