Abstract:HepatitisBvirus (HBV)exclusively infects liver parenchymal celsand formscovalently closedcircular DNA (cccDNA)withintheirnuclei.HBVcccDNAservesastheessntial templateforviralgenetranscription,thesolesourceofprogeny virus production,andthekeydriverofviralantigenexpresion,anditisthemolecularbasisforthepersistenceofHBVifection. Therefore,eliminationand/orfunctionalsilencingofccDNAis thekeytoeradicatechronicHBVinfection.Thisarticlediscuses thecriticalscientificissues thatneedtobesolvedduring eliminationoftheharmofHBVinfectionfromtheperspectivesof the synthesis,transcriptionandclearanceofccDA,aswellastheimpactofonparenchyalcelloncDA,inordertoprovide areferencefor eradicatingHBVinfectioninthefuture.
