Abstract:NBS-LRR is the largest gene family amongplantdiseaseresistance genes and playsanimportantrole in plantresistance and immuneresponse.Basedonthe previouslycompletedwhole-genomedataofDracaenacambodianaPiereexGagn.byourresearchgroup,thisstudysystematicallyidentifedthe NBS-LRR gene familyof D .cambodiana using bioinformatics methods,and analyzed itsphysicochemical properties,chromosome localization,gene structureandcodon preference.Theresults indicated thatthere wereatotal of 95 DeNBS-LRRcoding genesin D.cambodiana,whichwereunevenlydistributedon14outof2Ochromosomes.Thesegenesweredividedintosixsubfamiliesincluding CNL,CN,RNL,RN,NL,and N. The NBS-LRR protein of D . cambodiana was generally hydrophobic and was predicted to bedistributed inthenucleus,cytoplasm,cellmembraneandchloroplast.DcNBS-LRR proteinshadtenrelativelyconserved motifs,andmostofthemwere arangedinstableorder.Codonbiasanalysis showedthatDNBS-LRR preferredcodons ending in A/U,andthere were31highfrequencycodons,of which17wereconsidered tobeoptimalcodons.Parityrule2(PR2)-plot analysis,efective numberofcodon (ENC)-plotanalysisand neutral plotanalysisshowedthatthecodonusagepreferenceof Dc 1 NBS-LRR gene family was mainly influenced bynatural selection.Codon usage frequencyanalysis indicated that Saccharomyces cerevisiae,Nicotiana tabacum and Arabidopsis thalianacould allserveas ideal receptors for heterologous expressionof DcNBS LRR genes.Inaddition,the expression dynamicsof DcNBS-LRR genes inresponse to Fusariumoxysporum infection wereanalyzed.Theresultsshowedthattheexpresionofmost DeNBS-LRR geneswassuppresed intheearlystageandmiddlestageof infection(12h,1d,3d,and7d),whiletheexpresionwashigherinnon-infectionand3daysof infection.Tissudyisth firstto identify and analyze the characteristicsof theNBS-LRR gene familyin D cambodiana.The research resultswill provide clues for revealing the disease resistance mechanism of D .cambodiana and the molecular mechanism of Dragon’s blood formation,and lay the foundation for the subsequent in-depth analysis of the function of DcNBS-LRR.
