Isolation and whole genome sequence analysis of a Bacillus strain from Equus kiang

Abstract:ABacillus strain TEB-1 with biocontrol effcts was isolated from the feces of Equus kiang.This studyconducted whole-genome sequencingandmulti-dimensionalbioinformaticsanalysisof strain TEB-1basedonthird-generation sequencing technology.The sequencing datawere processedthrough gene assembly,geneprediction,andfunctionalannotation,within-depthminingandbiosyntheticpotentialevaluationofsecondarymetabolitesynthesisgeneclusters usingsoftware such asantiSMASH.The inhibitory effctsofstrain TEB-1against twoplantpathogenic fungi,Coletotrichum siamense and Corynespora cassicola,wereverified using the mycelial growthrate method,and drug resistance tests were performed on strain TEB-1.Theresultsshowedthat thecomplete genomeofstrain TEB-1was 4102747bp inlength,witha G+C content of 43.8% ,containing 4 O65 coding genes,85 tRNAs,and 3O rRNA genes.Bioinformatics analysis revealed abundantsecondary metabolitesynthesis geneclusters and multipledrug resistancegenes inthis strain.Functional validationresultsindicatedthatthesterilefermentationbrothofthestrainexhibitedvaryingdegreesofinhibitoryactivityagainstColletotrichum siamenseand Corynesporacassicolaat concentrations of 2.5% ,5.0%, 7.5% ,and 10.0% . Atthe 10.0% concentration,the inhibition rates reached 51.4% and 38.7% ,respectively.K-B disk diffusion tests demonstrated that strain TEB-1 was resistant to the antibioticsampicilln,penicilin,andlincomycin,whileshowinghighsensitivitytotetracycline,gentamicin,chloramphenicoleryt romycin,ciprofloxacin,cotrimoxazole,andceftraxone.TesefindingsindicatethatstrainTEB-1possessessignificantantifungalactivityalongsidecontrollbleresistancecharacteristics.Theresultsofthisstudyprovideatheoreticalbasisfordeveloping strain TEB-1 into an environmentally friendly biopesticide.
