Abstract:As a core indicator of cotton fiberquality, fiberlength plays a crucial role in accelerating the breding process through the development of itsmolecular markers.In thisstudy,amolecular marker(GhCRK26-1931 SNP)associatedwithcottonfiberlengthwasdevelopedvialocalization based on bulked segregant analysis combined with whole-genome sequencing(BSA-Seq),and itsapplication value was systematically evaluated.Competitiveallele-specific PCR(KASP)genotypingand phenotypic analysis were conducted on 512 coton varieties (lines).Theresultsshowed thatthe tested materialsexhibitedrichgeneticdiversity,andthereweresignificantphenotypicvariationsinfiberlengthamong extremefibermaterialsof Gossypium hirsutum-Gossypium barbadense backcrosspopulations, G. hirsutum germplasm materials,and G barbadense germplasm materials,with the coeficient of variation ranging from 3.48% to 9.65% . Cluster analysis based on phenotypic data suchas fiberlengthdividedthe materials intofour majorgroups.Verificationtestresults indicated thatthegenotypingresultsof GhCRK26-1931 SNP molecular marker in the parental Line 9 of the G∗ hirsutum-G. barbadense backcross populations and Xinhai 16 werecompletelyconsistentwiththeBSA-seqdata:Line9hadthe TTgenotype,while Xinhai16hadtheCC genotype.Among 60 extreme materials of the G* hirsutum-G. barbadense backcross populations,there was a significant difference in fiber length between the CC genotype(37 materials) and the TT genotype (23 materials)( P<0.05 ),and the average fiber length of materials withtheTTgenotypewasshorterthanthatofmaterialswiththeCC genotype.Furthermore,asimilarpaternwasobserved in305 G. hirsutummaterialsand145G.barbadensematerials:theaveragefiberlengthofmaterialswiththeTTgenotypewasstillsorter than that of materials with the CC genotype ( P<0.05 ). In conclusion,the GhCRK26-I931 SNP molecular marker can be effectivelyusedintheselectivebredingforcottonfiberlength,andtheCC genotypehasapositiveeffectonimprovingfiberength. This studynotonlyopens upanew avenueforthe breding of high-qualitycottonvarietiesbut alsoprovides areliable tol for marker-assisted selection of cotton fiber quality.
