Effect of EK2 Diluent on Sperm Motility and Artificial Insemination Outcomes in Linwu Ducks

Abstract: This study aimed to explore the effect of different dilution ratios of duck semen on artificial insemination. Semen was collected from Linwu ducks and diluted at ratios of 1 : 1 and 1 : 3 with EK2 diluent for artificial insemination. Post-dilution sperm motility, sperm viability, and sperm motility parameters [straight-line velocity (VSL), curvilinear velocity (VCL), and average path velocity (VAP)], fertility rate, and hatchability were assessed. The results showed that at the time point of 4 h post-dilution, the sperm motility, viability, and motility performance of both 1 : 1 and 1 : 3 dilution groups were extremely significantly higher than that in the normal saline dilution group. At the time point of 6 h post-dilution, sperm motility in the 1 : 3 dilution group was extremely significantly higher than that in the 1 : 1 dilution group. During 1–6 d post-insemination, the average fertility rates and hatchability of the 1 : 1 dilution, 1 : 3 dilution, and original semen groups had no significant differences. During 7–12 d, the average fertility rate of the original semen group was extremely significantly higher than those of the 1 : 1 and 1 : 3 dilution groups. Daily fertility rate analysis revealed that the 1 : 1 and 1 : 3 dilution groups had higher fertility rate than the original semen group during 1–4 d and had lower fertility rate during 5–12 d. In conclusion, ideal fertility can be achieved by inseminating semen diluted with EK2 diluent at a ratio of 1 : 3 or 1 : 1 (with an effective sperm count of no less than 1.0×108 ). Considering the fertility trend, it was recommended to perform artificial insemination every 4 d in production.
