Genetic Diversity Analysis of 16 Mulberry (Morus L.) Germplasm Accessions Based on ISSR Markers

Abstract: This study employed inter-simple sequence repeat (ISSR) markers to investigate the genetic diversity of 16 mulberry germplasm accessions from the mulberry germplasm nursery of Hunan Institute of Cotton and Sericulture. Seven effective ISSR primers were screened out, and amplification with these primers yielded a total of 73 bands, among which 60 were polymorphic bands, with the polymorphic loci accounting for 82.19%. The observed number of alleles, effective number of alleles, Nei's gene diversity index, and Shannon's information index were 1.821 9, 1.479 2, 0.280 0, and 0.420 5, respectively, which indicated rich genetic diversity of the mulberry germplasm accessions. The 16 mulberry germplasm accessions showed the genetic distances ranging from 0.042 0 to 0.764 1 and the genetic similarity coefficients ranging from 0.465 8 to 0.958 9. 'Yu 71-1 6' and 'Yu 71-1 7' showed the shortest genetic distance (0.042 0), with the closest genetic relationship. 'Yu 71-1 4' and 'Xiangsang 6' exhibited the longest genetic distance (0.764 1), which indicated the farthest genetic relationship. The results of UPGMA cluster analysis indicated that the 16 mulberry germplasm accessions could be assigned into five major groups at the genetic distance of 0.71.
