ZBTB7C regulates the proliferation and apoptosis of colorectal cancer cells by inhibiting aneuploid formation

ABSTRACT:To investigate the effects of ZBTB7C,which encodes a Zinc finger and BTB domain-containing protein linked totumor aneuploidy,on the proliferation and apoptosis of colorectal cancer cels,bioinformatic analyses Were applied to examine the corelation between ZBTB7C expresion and aneuploidy levels and prognosis in colorectal cancer tumor tissues. Subsequently,a ZBTB7C-knockdown HCT116celine wasconstructed using lentiviral infection.Chromosome spread and karyotype analysis were used to study the regulatory function of ZBTB7C on aneuploidy levels in HCT116 cels.The effectof ZBTB7Concellproliferation wasanalyzedusing livecellcounting andcolonyformation assays.The roleof ZBTB7C in cellapoptosis was studied through Western blot and Caspase-3 activity assays.Theeffectof ZBTB7Con in vivo tumor growth was investigated using a nude mouse subcutaneous xenograft model. The results showed that ZBTB7C knockdown significantly slowed the proliferation rate of HCT116cells.Aditionall,Caspase-3 activity was markedly enhanced,accompanied by increased expresion levels of cleaved PARPl and P21 and decreased expresion levels of BCL2.Furthermore,the weight and volume of the subcutaneous xenograft tumors in mice were significantly reduced upon ZBTB7C knockdown. In conclusion,the ZBTB7C gene may play a crucial regulatory role in the development and progression of colorectal cancer by inhibiting the production of aneuploid tumor cels.
