Correlation between triterpenoid synthesis and defense enzyme activities in Inonotus hispidus induced by oleic acid

ABSTRACT: Inonotus hispidus is a medicinal fungus whose main pharmacologicall active components are triterpenoids, which posess various biologicalactivities such asantioxidantandantitumor efects.However,under natural conditions, itsfruiting bodies exhibitalong growth cycle and lowtriterpenoidcontent,which limits itsmedicinal development potential.Toexplore methods forincreasing the yieldof triterpenoids in Inonotus hispidusandtheirregulatory mechanisms, this studyused oleic acid asan inducer,examined the activitychangesof key defense enzymes inthe mycelia,and analyzed their dynamic relationship with triterpenoid biosynthesis. The results showed that 0.4% oleic acid under liquid culture conditions significantly increased the total triterpenoid content,with the triterpenoid synthesis rate peaking at 84h and the total triterpenoid content reaching its maximum at 144h , Oleic acid activated the activities of intracellular defense enzymes,including superoxide dismutase (SOD),catalase(CAT),and peroxidase (POD).During the early stage of the stressresponse,increased SODactivities were strongly positivelycorrelated withthe triterpenoid synthesisrate.Asthe duction time was prolonged,the activities of thethreedefenseenzymesandthetriterpenoid synthesisrateshowedasignificantpositive corelation.These findings providea theoretical foundation for enhancing the biosynthesisof triterpenoid natural products.
