Fumigation Mechanism of Eugenol on Meloidogyne incognita

Abstract: Meloidogyne incognita isoneof the most destructive plant-parasitic nematodes (PPNs)in the world,which poses a serious threat to many important economic crops and causes huge agricultural economic losss every year. In this study,the plant-derived compound eugenol was used as the research object to exploreits mechanism offumigation against M incognita.The results of in vitro fumigation bioassay showed that eugenol had significant inhibitory activity against the second instar larvae (J2s) of M☉ incognita,and its LC50 value was 123.453μg/mL . Structural observation showed that after treatment with 800μg/mL eugenol for 24h ,thestructural integrity of J2s body wall was destroyed, which was manifested as body surface relaxation,bodyring enlargement, inclusion leakage and significant vacuolization,and abnormal expansion of esophageal gland.The physiological and biochemical analysis further revealed that eugenolcould cause body fluid extravasation and electrical conductivity increase of J2s and interfere with its osmotic regulation and respiratory metabolism proces.In addition,it also significantly afected the activityof many key enzymes in M. incognita J2s, including inhibiting detoxification function, disrupting antioxidant defense system and interfering with nerve signal transduction and causing nutrient metabolism disorders.The results showed that eugenol ultimately led to thedeath of nematodes bydestroying structural integrity,interfering with physiological metabolism and energy supply.The findings would provide atheoretical basis for the development of eugenol as an environmentally friendly fumigant.
