Genetic Diversity Analysis and DNA Fingerprint Construction of Dendrobium anosmum Population

Abstract: Using 40 Dendrobium anosmum germplasm resources from diferent regions as the subject,the iPBS molecular marker technique was employed to study their genetic diversityand population genetic diferentiation,and DNA fingerprinting Was constructed.Theresults show thata total of83clear bands were amplified using fiveiPBS primers, including 75 polymorphic bands,with a polymorphism rate of 87.95% , indicating that the selected primers had high eficiecy in detecting polymorphism.Genetic similarity analysis indicated that the genetic similarity coeficients among he 40 germplasms ranged from 0.5432 to 0.9703, with a mean of 0.7736 and a span of 0.4271,reflecting significant genetic variation among D . anosmum germplasms. Population genetic parameter analysis revealed that the observed number of alleles (Na) was 1.8795, the effective number of alleles (Ne) was 1.4642, the Nei's genetic diversity index (He) was O.2760,and the Shannon information index (I) was O.4183, further confirming the level of genetic diversity in the species.Based on the genetic similaritycoeficients,UPGMA cluster analysis indicated that when the similaritycoeficient reachedO.756,the40 germplasmscould beclearlydivided into six major groups,andthis classification result was significantly corelated with the geographical distribution characteristics of the germplasms.This study would provide important molecular biological evidence forthecollection and preservation, variety breeding,and sustainable utilization of D . anosmum germplasm resources.
