Screening of Interacting Proteins for HbERF1o9 in Rubber Tree (Hevea brasiliensis)

Abstract: The transcription factor ERF109,a member of the AP2/ERF subfamily,plays a pivotal role in plant stress responses.To further elucidate the molecular mechanism of HbERF109 in Hevea brasiliensis under low-temperature stress and exploreitspotential regulatorsin stress resistance,the structureof HbERF109 wascharacterized,its auto-activation activityeliminated through protein truncation,and interacting proteins were screened usinga yeast two-hybrid (Y2H) library. Candidate interactors were further validated by bimolecular fluorescence complementation (BiFC)assays.The ful-length HbERF109 exhibited strong auto-activation activity,which was abolishedafter truncation of the first 83 amino acids at the N-terminus (△N). Using the constructed bait vector pGBKT7-HbERF109-△N for library screning,a totalof30 potential interacting proteins,including HbSODand HbAPX, were identified.The proteins are functionallyimplicated inphotosynthesis,material transport,geneexpressonregulation,stressresponse,and disease defense.Yeast two-hybrid and BiFC assays further confirmed the interactions between HbERF109 and HbSOD as wellas HbAPX.Colectively,the findings indicate that HbERF109 mayfunction as an integrative regulatory component in the low-temperature stress response of rubber tree by coordinating a multi-layered protein interaction network associated with photosyntheticperformance,antioxidant capacity,and watertransport.This study would provide new insights into the molecular mechanism through which HbERF109 contributes to the stress resistance of Hevea brasiliensis.
