Abstract:This study investigates the effects of low-temperature preservation at 4∘C ontheviabilityofSalviamiltiorrhiza seed roots andcalls,andanalyzes therelationshipbetweenpreservation medium,rootsegment size,preservationduration and root germination rate,aiming to provide technical basis for the medium-and long-term preservation of S. miltiorrhiza germplasm resources. Using fresh root segments of different sizes and primary calli from one-year-old S. miltiorrhiza as materials,thisexperiment divides therootsegments into four groups basedonlengthanddiameter,determiningtheseedroot germinationrate bythesand culture method,and evaluating calus viabilitythrough survivalrateofcali during subculture. The results show that wet sand preservation at 4°C effectivelymaintains the seed root quality,significantly inhibits premature germination,preserves watercontent and viabilityand yields ahigh germinationrate.Root segment lengthanddiameter exertasignificant effecton sedling establishmentrate after low-temperature preservation.Most seed roots maintain a seedling establishment rate above 85% within one year of low-temperature preservation,and with increases in the length and diameter of the S.miltiorrhizaroot segments,both the sedling emergencerate and seedling height correspondinglyrise. Calli retain seedling formation capacity after 9~12 months of preservation at 4∘C ,although variations in preservation duration tolerance and viability exist among diferent varieties.Insummary,low-temperature wet sand preservationat 4∘C (204号 allows large-dimensionS.miltiorrhizaseedroots tobestoredforuptooneyear,providingafeasibleapproachtoreduce the costof biennialplanting.Additionally,Cali maintainstrong seedling formationcapacitywithin9monthsoflow-temperature preservation,offering a basis for reducing subculture frequency and simplifying the preservation protocols.
