MYB212 mediates SWEET11.3 to promote the sugar transport in Euryale ferox

ABSTRACT: SWEET proteins playa pivotal role in transporting sucrose,glucose,and fructose,particularly in phloem sucrose loading.To exploretheregulatoryroleof SWEETinE.ferox sugar transport,this studyutilizedthe‘PurpleflowerE.ferox’ cultivar as experimental material. Transcriptomic analysis of adult leaves and seed kernels identified EfSWEET11.3 as highly expressed insubmerged leaves and seed kernels,indicating its keyroleinsink tissuedevelopment.Phylogenetictreconstructionand motif analysis revealed that the EfSWEET1.3 protein contained eight conserved motifs,showing closest homology to Lotus(Nelumbo nucifera Gaertn.)and Nymphaea colorata(Nymphaea colorata).Sequence comparison and motif analysisconfirmed its structural similarity to hexose transporter genes.Bioinformatics and subcelular localizationassys demonstrated that EfSWEET11.3 possessed seven transmembrane domains,localizing to the cellmembrane as a bidirectional transmembrane transporter.Theexpresion trendofEfMYB2l2inthesed kernelsof differentstages of Euryale ferox was consistent with EfSWEET1.3,showing a significant positive corelation. Dual-luciferase assays further confirmed thatthe EfMYB212 transcription factor directly and specifically bound to the EfSWEET11.3promoter, enhancing its gene expression.This study provides a theoretical basis for understanding the regulatory mechanisms of EfMYB212 and EfSWEET11. 3 in E. ferox sugar transport pathways.
