Abstract : Betukladin is a dietary supplement composed of reindeer lichen powder and birch bark extract,possessing variouspharmacologicalactivitiessuchasbloodglucoseregulation,lipidlowering,anti-inflammatoryeffcts,and anticoagulation.This studyinvestigates thelipid-loweringactivityandunderlyingmechanismof Betukladin,aiming to validateits eficacyand provideascientific basis fordeveloping safeandeconomical naturalipid-lowering drugs.Using a zebrafishhyperlipidemiamodel,wefoundthatBetukladinsignificantlyimprovedtheelevatedOiRedOstainingintegrated opticaldensityvalues,triglyceride levels,andtotalcholesterol levels inducedbyhigh-fatmodeling,indicatingitslipidloweringactivity.Networkpharmacologyanalysissuggestedthat Betukladinmayexertitslipid-loweringeffectsthrough multiple coordinated pathways,including lipid and atherosclerosis pathways and the PPAR signaling pathway.RT-qPCR resultsshowedthatitcanregulate lipidmetabolism-related genes(apoAl,LDLR,andANGPTL3),fatyacidsynthesis and oxidation genes(SREBF1,FASN,PPARα,andcptla,),andcholesterol metabolism-related genes(CYP7A1, HMGCR,ABCG5,and ABCG8).
