Screening quality-difference markers of Qingxiao Wuwei Decoction
using an integrated“ fingerprint-pattern recognition-serum
migration components” strategy

Abstract: By establishing high-performance liquid chromatography(HPLC)fingerprints of Qingxiao Wuwei Decoction (QXWWD)and integrating chemical patern recognition with invivoand in vitrocomponent identificationmethods,this studyaimedtoscreen diferentialcompoundsand providereferencedataforconsistencyevaluationandthedevelopment of compoundpreparations.Thefingerprints of 18batches of QXWWDwere established evaluatedforsimilarity. Bioinformatics methods were used to analyze potential qualitymarkers.HPLC-Q-Exactive-MS was used to identify in vitro chemicalcomponentsand serummigrationcomponentsofQXwWD.Basedonthe integrated“fingerprint-pattern recognition-serum-migrant components”strategy,quality-diference markers werescreened.Atotal of 28peaks were identified inthefingerprintsofQXwWD,among which8quality-diferencemarkers,suchaschrysophanol,wereselected. Intotal,85invirochemicalcomponentsand55 serummigrationcomponentsof QXwWDwereidentified.Byfindingthe intersectionof thefingerprint-identifiedcomponentsandthoseidentifiedinvivoandinvitrousinga Venndiagram,we idenifiedchrysophanolasthecorequality-diferencemarker.Thisstudyestablishedasimpleandreproducibleanalytical method that can efectively evaluate thequality stabityand batch consistencyof QXwWD.This method provides reliable datato support processoptimization,quality-standarddevelopment,andthesubsequent developmentof this prescription.
