Study on Response Surface Methodology Optimization of Pigment Extraction from Cotton Stem Bark and Its Antioxidant Activity

Abstract:Tooptimizethe extraction process ofnatural pigments fromcoton stalk fibers andanalyze their antioxidant activity, the 1,1 -diphenyl-2-picrylhydrazyl(DPPH)radical scavenging activity,2,2'-azino-bis 3-ethylbenzothiazoline-6- sulfonic acid(ABTS)radical scavenging abilityand iron-reducing power were taken as the key indicators.A strategy combining ultrasonic-asisted technologyand response surface methodology was employed to optimize the extraction conditions.Theresults showed thattheoptimalpigment extractionconditions wereas follows:liquid-to-materialratio of 104.036 mL·g-1 ,ethanol content of 73.462% ,ultrasonic temperature of 59.801°C ,and extraction time of 100.056 min.Under these conditions,the extracted pigment content was 51.646mg⋅g-1 .Theantioxidant testindicated that this pigment hadastrong scavenging ability for DPPH,ABTS free radicalsand iron ions,and the half-maximal inhibitory concentration( IC50 )forDPPH was 46.201 39 μo⋅mL-1 ,while forABTS the IO 50 was 19.542 30μg⋅mL-1 : The iron-reducing assay test showed that at a pigment content of 1mg⋅mL-1 ,its activity was almost equivalent to that of vitamin C.Aboveresultsopenednewavenues forthecomprehensiveutilizationofcotton,particularly inresourceeffcient applications and food additive development,showing great potential for future application.
