Abstract:Cottonfiberlength directlyaffects thequalityyarnfabric,whichisakey indicatorfor evaluating the quality economic value fiber quality.The weighted gene co-expression network analysis(WGCNA) identifies coordinated geneexpresion patterns toreveal potential associations between keyregulatory genes traits, facilitating a deeper understing gene regulatory networks.Using the transcriptome data from 4coton cultivars (2 with long fibers 2 with medium fibers)across key stages fiber development(O~3Od post-anthesis),the WGCNAwereperformed,4modules were identified tobesignificantly positively correlatedwith fiber development,including turquoise,blue,brown yelow modules,respectively.Among them,the blue module showed high association with fiber length during therapid elongation phase(1O~15d post-anthesis).GO(genet ontology) KEGG(Kyoto encyclopedia genes genomes)enrichment analyses revealed that genes the blue module were mainly involved in protein transmembrane transport.Further analysis module membership (MM) gene significance(GS)identified GhM_AO8G1439 as a cidate gene encoding amember the tetraspanin protein.Subsequently,29 members Tetraspanin genefamily inthe genomeuplcotton were identified.Based on their chromosomal physical positions,the GhM_AO8G1439 was named GhTET8.The tissue-specific expression analysis showed thatGhTET8was highlyexpressd in fiber,petals roos.Subcellular localizationanalysis revealedthat GhTET8 protein was localized totheplasma membrane.Functional studies demonstrated that overexpression GhTET8significantly promoted trichome elongation inArabidopsis thaliana,whereas silencing this geneincotonledtosignificantreduction infiberlength.Colectively,GhTET8 positivelyregulatedfiberelongation, which laid theoretical basis forits application in coton molecular breeding programs to enhance fiber quality.
