Evaluation of Nematicidal Activity of Coumarin Compounds Against Meloidogyne incognita

Abstract: Meloidogyne incognita is one of the plant obligate parasitic pathogenic nematodes, which is seriously harmful to tomato and tobacco. In recent years,with the increase of pesticides application, the resistance of Meloidogyne incognita to pesticides has gradually increased, and it is necessary to screen new substitute agents as alternative nematicides. This study evaluated the bioactivity of 1l common coumarin compounds against M . incognita through bioassays. Root galls from tobacco roots were excised and incubated to prepare a nematode suspension at a concentration of 5O individuals/ 100μL . The number of dead nematodes treated with different compounds was microscopically examined in 96-well plates after 24h and 48h of treatments.Bioassay results showed that coumarin and 6-methylcoumarin exhibited excellent bioactivity against M . incognita at 125mg/L ,with 24hLC50 value of 19.22mg/L and 39.44mg/L ,respectively. In the field experiment, Treat Cl significantly increased the plant height and maximum leaf length during the vigorous growth stage of plants,as wellas the plant height, stem diameter,and number of effective leaves during the topping stage. At the end of Treat Cl and Treat C3 had the lowest disease index (23.69 and 25.48,respectively),while the control group had a disease index of 41.83. The corresponding control efficiency was 43.37% and 39.09% ,respectively. In this experiment,coumarin and 6-methylcoumarin were screened out as safe and efficient nematicides,with good application prospects.
