基于网络药理学及分子对接技术探究清热利湿方治疗急性痛风性关节炎作用机制

【Abstract】Objective:To explore the action mechanism of the heat-dampness clearing prescription in the treatmentof acute gouty arthritis(AGA) through network pharmacology and molecular docking technology.Methods::The active components and related targets of this prescription were screened through the TCMSP platform.Disease-related targets were screened through databases such as GeneCards,TTD, OMIM,and DRUGBANK. After mapping the target components of the prescription and the disease-related targets of AGA using R language (eulerrpackage),the intersectiontargets of herbs and diseases were obtained.567 disease-related targets were obtainedandthe intersection targets of the prescription components and AGA were 6O,and.The network diagram of the prescription components -intersection targets-signaling pathways was constructed using Cytoscape 3.9.O software.Finally,the core targets and maincomponents were selected for molecular docking using PyMOL2.4.O software.Results:8O potential active componentsand 259 target sitesof the prescription were screened out.567 disease-related targets and 6O intersection targets of the prescription components and diseases were obtained.GO and KEGG enrichment analysis revealed that the intersection targets were mainlyinvolved inbiological processessuchasinflammatoryresponseandregulationof inflammatory response,and involved NOD-like receptors,Toll-like receptors,NF-kB,IL-17 signaling pathways,etc. Molecular docking results showed that most of the main active components of the herbal medicine could bind well to the core targets (binding energy ⩽-5.0kcal/mol, . Conclusion: The heat - dampness clearing prescription acts on multiple components and multiple pathways to improve AGA,providing ascientific basis forthe clinical application of the prescription.
