【Abstract】Objective:To explore the mechanism of Portulacas in the treatment of ulcerative colitis (UC)and to establishacomprehensiveand optimized network pharmacologystrategy,soas toefectively compensate for the deficiencyof existing network pharmacology in ignoring the synergistic mechanism of Portulacas of carbohydrates,organic acids and flavonoids inthe treatment ofUC.Methods:ThroughLC-MSandacomprehensive optimized network pharmacologyapproach,the synergistic mechanismof the three types ofcomponents in Portulacas inthe treatmentof ulcerativecolitis was comprehensively investigated.Firstly,LC-MS technology was used to identifytheactivecomponents inPortulacas.Subsequently,data mining and a comprehensive network pharmacology strategy wereemployed tocomprehensively explore the efects and potential mechanisms of the measured components and a few reported Portulacas carbohydrates and organicacids inthe treatment of UC.Results:Thecarbohydratesand organicacids in Portulacas have important synergistic efects with flavonoids in the treatment of UC.Thecomprehensive network pharmacology strategy predicted that glucuronic acid(GlcA),rhamnose(RH3),and mannose(MAN)exert their efects byregulating STAT3 protein.Luteolin,kaempferol and quercetin mainly aect SRC,AKT1and PIK3Rl proteins whilep-coumaric acid(PCA)and cafeic acid(CAA)mainly exert their efects through PIK3CA and ESR1 proteins.Cancer pathways and PI3K -Akt signaling pathway were the key pathways for Portulacas in the treatment of UC.Conclusion: Theflavonoids in Portulacas have a prominent affnity for inflammatory protein molecules.GlcA,RH3,MANand PCA,CAA have been proven to play a core role in the synergistic treatment of UC.
