ABSTRACTOBJECTIVEToanalyze the prototype components absorbed into bloodand brainofLithocarpuslitseifolius leaves, soas toprovideareferenceforclarifyingthepharmacological materialbasisof itspreventionandtreatmentofcentralnerous systemdiseases.METHODSTheethanolextractofL.litseifoliusleaves,aswellasthegastriclavagefluidandperfusionsolution wereprepared.Usingratsassubjects,plasmasamplesof intestinal wall metabolism,intestinal florametabolismandhepatic metabolismwerepreparedviainsituintestinalperfusionandclosed intestinallopmethod;whilecomprehensive metabolicplasma samples,braintissesamples,andcerebrospinalfluidsampleswerecolectedafterintragastricadministration.UPLC-HRMS technologywasutlizedtoanalyzeandidentifychemicalcomponentsandprototypecomponentsabsorbedintobloodandbrainof L , litseifoliusleaves.RESULTSAtotalof66chemicalconstituentswereidentifiedinL.litseifoliusleaves,primarilyconsistingof flavonoids,organicacids,andothers.Atotalof16,13,11,and5prototypecomponents wereidentifiedinintestinal wal metabolism,intestinal florametabolism,hepaticmetabolism,andcomprehensivemetabolicplasmasamples,respectively. Additioall,4prototeomponentseredetectedinbraintisseand9incerebrospialfluidlorzin,rlbatinoinO -malonylhexoside,and phloretinwereidentifiedascommoncomponentsacross allsample types.CONCLUsIONsPrototype components absorbed into blood and brain of L .litseifolius leaves,such as phloridzin,trilobatin,phloretin,and other components may serve as the pharmacological material basis for their therapeutic efects on central nervous system diseases.
