Optimization of high-density fermentation process for Bacillus velezensis G127

Abstract Bacillus velezensis is widely distributed and has high biological activity,which is a safe and reliable biocontrol strain.Reducing cost and increasing cellbiomassis the key to its wide application.The optimal carbon source,nitrogen source and inorganic salt were selected by single factor experiment,and then the high-density fermentation conditions and process of Bacillus velezensis Gl27 were optimized by response surface. The optimal medium composition and culture conditions were as follows:beef paste supplemental level 15.2g/L ,molasses 13.0g/L ,sodium chloride 5.0g/L ,fermentation time 72h ,fermentation temperature 30∘C ,inoculation amount 3.3% ,and initial pH value 5.1. The optimized biomass could reach 5.45×1010 CFU/mL. The obtained high-density fermentation technology provides guarantee for the large-scale production and application of Bacillus velezensis G127.
